Detection of Fermentative Variants with Tetrazolium.

چکیده

While trying to develop methods for finding dehydrogenase mutants, we found tetrazolium (triphenyl tetrazolium chloride) to be useful for detecting fermentative variants. This indicator has the advantage that the nonfermenting variants are stained a deep red whereas the fermenters and the background are neutral. For example, cultures of Escherichia coli K-12 were spread on nutrient agar plates containing 1 per cent carbohydrate and 0.005 per cent tetrazolium. The plates were then irradiated with an ultraviolet lamp long enough to leave 100 to 500 colonies per plate after incubation. Red colonies or sectors were transferred to EMB agar to purify and verify them as mutants. Yields, ranging from 0.1 per cent to 0.01 per cent of the survivors, of mutants unable to ferment lactose, maltose, or glucose, respectively, have been obtained. The response is undoubtedly due to the pH dependence of t’he biological reduction of tetrazolium to the highly colored, insoluble formazan. This usually accumulates in a single large granule near one pole of the cell. Since Salmonella and Shigella cultures show a strong red reaction on lactose tetrazolium agar, this medium might be developed as an indicator for the enteric pathogens. However, it will be necessary to eliminate the reactions given hy weak lactose fermenters (by cont’rolling the pH and buffer capacity of the medium) and probably also to incorporate a selective inhibitor such as brilliant green.

Detection of rough dissociants of Pasteurella pestis with tetrazolium chloride.

Lederberg (J. Bact., 56, 695, 1948) reported the use of triphenyl tetrazolium chloride to detect fermentative variants of Ewherichia coli K-12. We have found that smooth and rough colonies of Pasteurella pestis could be identified easily when grown on tryptose (Difco) agar containing this compound. On many routine and modified laboratory media that did not contain tetrazolium, differentiation w...

Detection of Metallo-Beta-lactamase Production in Rare Carbapenem-Resistant Non-fermentative Gram-Negative Bacilli Isolated in a Tertiary Care Hospital, Visakhapatnam, India

Introduction: Non-fermentative Gram-negative bacilli (NFGNB) are occasionally involved in infectious diseases pathology, but have shown resistance to multiple antibiotics and the capability to gain new resistance factors in the hospital environment. The present study was aimed to investigate the antibiotic susceptibility pattern of rare NFGNB isolated from different clinical samples and the pre...

Concept drift detection in event logs using statistical information of variants

In recent years, business process management (BPM) has been highly regarded as an improvement in the efficiency and effectiveness of organizations. Extracting and analyzing information on business processes is an important part of this structure. But these processes are not sustainable over time and may change for a variety of reasons, such as the environment and human resources. These changes ...

A fast and easy nitroblue tetrazolium method for carrier screening and prenatal detection of chronic granulomatous disease.

BACKGROUND Analysis of the functional activity of neutrophils is of great importance in the differential diagnosis of patients with recurrent bacterial infections. It has long been established that stimulated polymorphonuclear leukocytes reduce nitroblue tetrazolium. Application of a simple and reliable nitroblue tetrazolium method that clearly differentiates the chronic granulomatous disease p...

Molecular Detection of Novel Genetic Variants Associated to Anaplasma ovis among Dromedary Camels in Iran

To the best of our knowledge, little information is available regarding the presence of Anaplasma species in camels in Iran. This study sought to investigate the presence of Anaplasma species by microscopy and polymerase chain reaction (PCR) assays in 100 healthy dromedaries (Camelus dromedarius) arriving for slaughter. The microscopic examination of Giemsa-stained blood films revealed that Ana...
